For centuries the world has been under constant threat by the forces of darkness, lead by the Yokai King, Ankokushi. But even while he has tried to spread his evil across the world, a long standing clan of hunters have always stopped him. The Shindai clan and their legendary weapon, the Yokai Sealer, has defeated the king of darkness and his many general countless times. His last defeat happened 70 years ago at the hands of Takemoto Kuro and his partner, Miyura Akatsuki. But even after this defeat, Ankokushi's loyal servant, Osorero, plans to resurrect his master once more. To prevent himself from being found, he has remained hidden in the shadows for years until he could find the right opportunity. But while he plotted, Ankokushi's eternal enemy Akira Yoakerin or Kuromoden, a Yokai turned against his own kind due to his love for the Earth after his witness to how humans protected each other, began keeping an eye on Death as to prevent him from resurrecting his master or the castle.

During the present day, Akira has tracked down Osorero to the city of Kyoto and begun preparing his own operations. Having kept in contact with the current descendant families of the Belmonts, Akira arranged for the new members to be sent to the city and begin their new training under his supervision. With help from many different groups such as the Kamizure Clan, Akuma Forgemasters, and several other hunter groups, they succeeded in creating new items called the Glyph stones to aid them in their battles.

A long time ago, a prophecy was made about the Hikari family that within centuries from their time, a young girl would be born as half human and half Yokai, and through difficulties that her Yokai half would exploit, especially giving Ankokushi an advantage to control the girl, she would lead her generation of slayers against the villainous Yokai King and destroy him and his followers once and for all.

In the present day within Kyoto, a young girl in her late teens is summoned to an old bookshop and cafe, riding a motorcycle with blood red decals. This girl is Akari Chinoko, descendant of the Hikari family and goddaughter to Akira Yoakerin/Kuromoden. As the newly designated leader of the latest generation of Slayerman, she will stop at nothing to bring Ankokushi down for good, even if it means hiding her Yokai nature from the other team members.
